Do I bypass the plugin when A/Bing with my monitors?

Hello, real rookie question here

It says in the user manual to A/B the monitor plugin with my real speakers, and change the settings until they sound the same. If the plugin is on the mix bus, do I need to bypass it whenever I check the speakers? It makes sesne to me but the manual doesn't say.
